Bhumi was born on 18 July 1989 to father late Mr. Satish Pednekar and mother Sumitra Hooda Pednekar in Mumbai, Maharashtra, India. Her father died on March 24, 2011. She has a younger sister Samiksha Pednekar who is a lawyer. Two sisters are the carbon copy of each other.
Pednekar did her schooling from Arya Vidya Mandir in Juhu, Mumbai where celebrities like Aishwarya Rai Bachchan, Arjun Kapoor, Karan Tacker, Kunal Deshmukh, Prateik Babbar, Rahul Bose, Sonakshi Sinha, and Sonam Kapoor studied. Then she went to Whistling Woods International, Mumbai to study Communication & media arts but expelled for poor attendance. Finally, she got her bachelor of commerce (B.Com) from University of Mumbai, Mumbai.

Since the age of 12, she wanted to be an actress. So that when she saw a vacancy post in Yash Raj Films she applied for the job. Before debuting in her acting career, she worked as an assistant casting director under Shanoo Sharma for Yash Raj Films for six years. Later in 2015, she made her acting debut with director Sharat Katariya‘s comedy-drama film ‘Dum Laga Ke Haisha‘ in 2015, opposite Ayushmann Khurrana which was produced by Aditya Chopra under Yash Raj Films banner. ‘Dum Laga Ke Haisha’ collects more than ₹30 crore from the budget of only ₹9 crore. Reportedly, to play an overweight woman character in the film, she gained almost 15 kg. She earned several ‘Best Female Debut‘ awards in several award shows including Filmfare Awards for the role.
The same year she appeared in Vikram Gupta directed Y-Films’s mini web-series ‘Man’s world‘ with a very small role.
In 2017, Pednekar appeared in debutante Shree Narayan Singh’s directorial comedy-drama film ‘Toilet: Ek Prem Katha’, opposite superstar Akshay Kumar. The film becomes one of the commercially successful films with worldwide earnings of over ₹300 crore from the budget of estimated ₹50 crore. The same year, she again appeared opposite Ayushmann Khurrana in R.S. Prasanna directed comedy-drama film ‘Shubh Mangal Saavdhan‘. That film again gave her huge fame and success with a collection of ₹65 crore at the box office from ₹25 crore budget.
